Collection Kit

Every Kit Box issued to its clients by Cryoviva is UN3373 certified and contains all the important contents required for collecting the Umbilical Cord Blood, Umbilical Cord Tissue & maternal blood sample. The collection kit contents are:

• Sterile bag for cord blood collection prefilled with anticoagulant and with a needle protector that helps in preventing accidental pricks.
• Absorbent pad, alcohol swabs, maternal blood collection vacutainer, povidone iodine prep-pad, sterile plastic cord clamp, parafilm, blood collection needle with luer lock adapter, vacutainer holder and tourniquet, a pair of sterile surgical gloves, gel pack etc.
• An insulated aluminium foil pouch for the collected cord blood bag which act as shock absorbers as well as long term temperature maintenance thus maintaining the quality of the product. 
• Two Falcon brand tubes, one for initial decontamination of umbilical cord & the second tube for collection & transportation of umbilical cord tissue to facility.
• The cord blood, cord tissue & maternal blood sample shall be further put in a secondary leak proof zip lock containers as per the IATA requirement.

Transportation container

The cell quality and viability of the collected cord blood sample is maintained optimally during the transportation with the help of UN3373 certified kit boxes. The kit box is carefully designed to ensure that the optimum temperature as per the regulatory & accreditation requirements is maintained during the transportation of collected samples. The kit box is made up of Thermocol brand fabric with outer hard cardboard & inner Thermocol container (Primary & Secondary container) packing as per the international IATA guidelines. The twin frozen gel packs are used to sandwich the collected cord blood bag, which in turn is packed using a 95 KPA bag, followed by an insulated aluminium foil pouch which act as a shock absorber as well as ensures the optimum temperature. The client receives an acknowledgement email that includes safely processing & storage of their sample in our facility.

Shipping Info

In the event that the cord blood cells are needed for therapy, Cryoviva has procedures to release and ship the sample. Cryoviva requires receipt of written request from the client and also a written requisition from the treating physician. Cryoviva will put in its best efforts to complete the documentation formalities for releasing the product as per the current regulatory norms in co-ordination with the treating physician. Cryoviva shall ship the product/s in an appropriate manner, such as use of controlled monitored cryo-shipper (temperature at or below minus 150°C) throughout the shipment process, in order to provide safe and timely transportation. Cryoviva would offer free shipment worldwide for the product as confirmed during the time of retrieval by the physician.

Processing Method

Purity begins at child birth & so does the preservation of life saving stem cells. It has always been an endeavour of Cryoviva to provide the best technology; hence Cryoviva was the first to bring automated cord blood processing technology to India in 2009. Currently in 2023 Cryoviva India relies on AXP® II System of automated processing technology. With automated technology each cord blood unit is fully processed in only 30-40 minutes. Cryoviva India follows the “Closed bag” processing protocol under the aseptic environment of laminar flow biological safety cabinets for harvesting cord blood stem cells which greatly reduces the possibility of microbial contamination.

Components Stored

At Cryoviva every cord blood sample is processed as per the best international standards and the following parameters are checked at the time of birth. Details of the test results are provided to each client of Cryoviva in their preservation report.

Maternal Blood Sample: Infectious diseases tests like HIV, Hepatitis B, Hepatitis C, HTLV, CMV, Syphilis, Malaria Infections & NAT (HIV1 RNA, HBV DNA & HCV RNA) are performed at the time of processing and storage. Additional maternal serum & plasma are also preserved for future testing purposes, if any.

Cord Blood Sample: ABO grouping and RH typing, Total Nucleated Cell (TNC) count, Mononuclear Cell (MNC) count, CD34 cell count, cell viability, sterility testing for microbial culture, microbial contamination, Colony Forming Unit (CFU) assay & Hemoglobinopathies.

Cord Tissue: Characterization report (both positive & negative), viability test, sterility, mycoplasma analysis, differentiation potential, endotoxin test, positive surface markers like CD 105, CD73, CD44 & CD90, negative surface markers like CD45, CD34, CD14, CD19 & HLA-DR, characteristics of differentiation into Osteoblasts, Adipocytes & Chondroblasts. All these tests are performed at the time of retrieval upon approval from respective government bodies in India and abroad as per the regulatory protocol of that country.

Additional Testing: HLA typing is performed on the 2^nd birth anniversary of the child by Next Generation Sequencing (NGS). If the cord blood unit is retrieved for therapy additional tests are performed at no cost to the client: HLA typing for both donor & recipient by NGS, TNC count, MNC count, sterility test, and viability test.

Storage Method

Cord Blood: The processed Cord Blood is preserved in a FDA-approved multi-compartment cryobag encased between specially designed aluminium cassettes. It is cooled by the standard process of controlled rate freezing and stored in the vapour phase of cryogenic nitrogen until reaching a temperature below -150 degree Centigrade.

Cord Tissue: At Cryoviva, are processed in a WHO-GMP complaint lab using a patented technology of Da Vinci Biosciences, USA. Cryoviva uses explants cell culture method and the cultured MSC will be released as per the current regulatory norms. 

Annual Stability Report: Every year Cryoviva shares an annual stability report to its clients, which shall contain contains statistical details of a donated sample which was stored in the same storage tank of the client. The donated sample is thawed & then tested for sterility, viability, CD34+ count, recovery percentage. Then the report is mapped with the pre-freeze report to check the stability of the specimen. 

Storage Duration: Cryoviva offers clients the option of either a standard storage plan for 21 years or a lifetime plan for 75 years from the birth anniversary of child.

Clinical Experience
